Ruth was born in Exeter House, Roehampton, Surrey, London. She was the seventh child of Richard Du Cane (1821-1904) and Charlotte Maria Guest (1834-1902). In her adult life she was described as being almost 6 feet tall. In January 1900, when Ruth expressed the desire to be trained in typing and shorthand, her maternal aunt Lady Enid Layard told her to attend Miss Oswell's secretarial school and she would pay for it. By June 1901, Ruth was living in London and working as a secretary for the Anti Lead Poisoning League. Ruth never married.
Ruth died of heart failure at 34 Delamere Terrace, Paddington, London on 16 May 1907 after a sixteen day illness of pneumonia. Her brother Richard Dacre Guest Du Cane was with her at the time of death. Ruth was the first of the Du Cane siblings to die and her remains were interred with her father Richard's remains. Her sister Mary "Daisy" Virginia was the next sibling to die (1919) and her remains were also interred in the grave plot.
